Involvement of S100A8 and S100A9 in nonischaemic cardiomyopathy

1. S100 calcium binding proteins A8 and A9 (S100A8/A9) could be released from bone marrow (BM)- and non-BM-derived cells.
2. S100A8/A9 mainly activates the Toll-like receptor 4 (TLR4) and receptors of advanced glycation endproducts (RAGE) signalling pathways and drives mitochondrial dysfunction, inflammation, reactive oxygen species production and cardiac hypertrophy.
3. Therapeutic inhibitors targeting S100A8/A9 (ABR-238901, paquinimod) or NLRP3 (CY-09) may be effective interventions.

Structural insights into retinoic acid receptor activation and selective modulators

1. Selective retinoic acid receptor (RAR) modulators regulate the assembly of the RAR transcription complex, which is composed of RAR, retinoic X receptor, modulators and co-regulators.
2. The RAR transcription complex recognises specific retinoic acid response elements, modulating the transcription of target genes.
3. The modulation influences differentiation, tumourigenesis and cancer metastasis, and also demonstrates potential efficacy in cancer treatment.
