Our cover features an immunofluorescent staining of a spleen section from a mouse which had been immunized with ovalbumin. The white pulp comprises CD3ɛ-positive T cells (red) and IgM-positive follicular B cells (blue). This is surrounded by a ring of MAdCAM-1-positive sinus-lining cells (green), followed by an outer layer of IgM-positive marginal zone B cells (blue). The image was provided by Preite et al., (pp. 3010–3021) who show that Cd3e-deficient mice, which lack T follicular helper (Tfh) cells, have germinal centre B cells with fewer somatic mutations as compared with wild-type mice, and which exhibit impaired affinity maturation and reduced differentiation into plasma cells. These phenomena could be reversed with reconstitution of Cd3e-deficient mice with wild-type Treg cells, demonstrating that regulated Tfh-cell responses are crucial to develop serological memory.
