Developing and Implementing a Colorectal Cancer Screening Program in Uganda: Stakeholder Perceived Barriers and Opportunities

Qualitative semi-structured interviews were held with stakeholders in Uganda with responsibilities for delivering cancer prevention, screening and policy/programming to elicit their views and improve understanding regarding barriers to, and opportunities for, introducing a colorectal cancer screening program. Results indicated significant barriers across policy, health system, community and individual levels whilst noting opportunities to overcome these challenges, such as the drafting of the first national cancer control plan. Key priorities include colorectal cancer primary prevention (e.g., mass awareness raising) and using implementation science to embed evidence-informed colorectal cancer screening in Uganda.

Addition of chemotherapy to standard radiation improved OS in women with LACC, regardless of HIV status. Our findings add to a body of literature highlighting the importance of providing concurrent chemoradiotherapy to all patients with LACC, including persons living with HIV and those with stage III/IV disease.
Biological Activity of Biomarkers Associated With Metastasis in Osteosarcoma Cell Lines

Osteosarcoma presents diagnostic and treatment challenges due to its unclear etiology and frequent late-stage detection. This study aims to identify metastasis biomarkers in osteosarcoma cell lines and biopsies. Through RNA evaluation, a gene functional enrichment analysis of DEGs enabled the identification of interaction networks in osteosarcoma, revealing potential biomarkers. Biochemical analysis elucidates the co-participation of TTX-sensitive NaVs and MMP-2 in facilitating migration and invasion, suggesting their suitability as novel prognostic biomarkers. Additionally, this study introduces a model delineating the potential interaction mechanism among ion channels, MMP-2, and other crucial factors in the metastatic cascade of osteosarcoma.

Real-World Outcomes of Newly Diagnosed Multiple Myeloma Patients Treated Before the Era of Anti-CD38 Antibodies: The EMMY Cohort From 2017 to 2020

A total of 1036 non-transplant eligible (NTE) patients and 561 patients who received autologous stem cell transplantation (ASCT) were enrolled in Emmy. In the ASCT group, patients treated with and without R maintenance had a mPFS of 51.8 (95% CI: 44.1–NA) and 29.6 months (95% CI: 21.8–40.9), respectively. In the NTE group, the mPFS was 26.3 (95% CI: 21.9–30.9) and 14.6 months (95% CI: 11.9–17.7) in patients who received an R-based and non-R-based regimen, respectively. The 2017–2020 period was marked by the expansion of R use in both NMDD ASCT and NTE patients.

In this study, we demonstrated that METL16 is essential for GC progression by connecting m6A modification with histone modifications. The regulatory role of METTL16, which suppresses the malignant phenotype of GC cells at both the transcriptional and post-transcriptional levels, offers new prospects for tumor diagnosis and precision treatment.

This study investigates the potential of radiomics to improve risk stratification for locoregional recurrence (LRR) in nasopharyngeal carcinoma (NPC) patients. By combining radiomic features with clinical data, the authors achieved improved prediction of LRR compared to using either alone. The most important radiomics feature was tumour size, suggesting a need to reconsider its role in NPC staging.

MA penetration at the urology practice-level varies considerably. Higher MA penetration was associated with increased treatment, namely surgery, among patients with Traditional Medicare without changes in quality. Our work suggests that, in the context of prostate cancer, increasing MA penetration may not always be associated with constrained utilization among Traditional Medicare beneficiaries.

Landscape of TET2 Mutations: From Hematological Malignancies to Solid Tumors

Somatic TET2 mutations are reported in numerous solid human cancers, including those arising in the skin, lung, and prostate, although there is a paucity of data on their frequency in these settings compared to hematological malignancies. TET2 mutations affect prognosis in myeloid leukemia and other hematopoietic malignancies, but little is known about their impact on solid tumors. Here, we review the data on TET2 mutation in human solid cancers and explore their impact on prognosis and patient outcome.

The Influence of Performance Status, Inflammation, and Nutrition on the Impact of SGLT2 Inhibitors on Cancer Outcomes

This study highlights the critical role of patients‘ baseline characteristics, including performance status, inflammatory markers and nutritional status, in determining cancer mortality or progression. Our results showed both a nonsignificant effect of SGLT2 inhibitors on cancer mortality or progression and a nonsignificant effect of SGLT2 inhibitors on performance status, inflammatory markers, and malnutrition. These findings suggest that the potential benefits of SGLT2 inhibitors in cancer treatment may be constrained under standard clinical dosing conditions.

This study uncovers dysregulated lipid metabolism pathways in thyroid cancer (TC), identifying six lipid metabolism-related genes (LMGs) as prognostic markers: FABP4, PPARGC1A, AGPAT4, ALDH1A1, GPAT3, and TGFA. A prognostic risk model based on these LMGs demonstrates superior predictive performance for overall survival. Dysregulated pathways, including the TCA cycle and glycerophospholipid metabolism, are implicated in TC progression. The risk model correlates with immune cell infiltration and chemotherapy sensitivity, guiding personalized therapeutic strategies. These findings highlight the potential of targeting dysregulated lipid metabolism pathways and LMGs for novel therapeutic interventions in TC.
Distress and Its Determinants in 820 Consecutive Melanoma Patients

Psycho-oncological distress is common in melanoma patients, with over 40% showing elevated distress levels (DT ≥ 5), particularly among women, younger individuals, and those with advanced disease stages (III–IV). Key complaints identified through the NCCN Problem List include fear, sleep disturbances, physical symptoms, and existential concerns. These findings emphasize the importance of targeted psycho-oncological support, especially for high-risk patient groups.

Using bioelectrical impedance measurements from the UK Biobank, we have shown that the allometric fat-mass and lean-mass indices (reflecting general obesity), as well as diabetes, were associated positively with pancreatic cancer risk, independent of each other and other known risk factors. No independent association of abdominal obesity with pancreatic cancer risk remained after factoring out the associations of waist size with general obesity and height and accounting for smoking and diabetes.

Perioperative Trends in Distress Among Cancer Patients: A Systematic Review and Meta-Analysis

Distress is common among cancer patients. A systematic review of 57 studies found that distress levels are moderate to severe preoperatively and persist through the early postoperative phase, gradually declining from the 1-month postoperative mark onwards. Notably, patients with brain cancer reported the highest postoperative distress levels, followed by those with breast cancer.

Real-Life Cohort of Patients With Resected High-Risk Melanoma Treated by Adjuvant Anti-PD1 Therapy

This real-life study based on 235 patients treated with adjuvant anti-PD1 therapy provides a comprehensive view of the outcomes and factors associated with recurrence in patients with resected high-risk stage III/IV melanoma. Ulceration emerged as the only significant predictive factor for recurrence and was associated with a decrease in recurrence-free survival at 24 months.
