Collagen fiber and cellular dynamics of axolotl skin with aging

Ambystoma mexicanum (axolotl) hasrelatively transparent skin, providing an ideal experimental environment forskin biology. However, their skin becomes opaque as they age. This studyfocuses on age-related changes in axolotl skin, including decreased numbers of collagen-producing fibroblasts, altered localization of type III collagen, changes in the thickness of dermal collagen layers, and alterations in fiberpatterns.

Efficient CRISPR/Cas9-mediated knockin of reporter genes in rats at ROSA26 by pronuclear microinjection

We optimized a CRISPR-mediated genome editing method for generating knockin (KI) rats through zygote microinjection, covering zygote harvesting to timed microinjection, which consistently yielded KI rats. Additionally, we established four fluorescent reporter lines at the ROSA26 locus. Our study includes detailed protocols for donor vector design, zygote collection, microinjection, founder screening, and cryopreservation in rats.
