Pladienolide, 12-gliedrige Makrolide aus Streptomyces platensis Mer-11107, zeigen herausragende In-vitro- und In-vivo-Antitumoraktivitäten auf der Basis eines einzigartigen Wirkmodus. Y. Kotake et al. beschreiben in der Zuschrift auf S. 4428 ff. die erste Totalsynthese dieser Naturstoffklasse über eine effiziente Kupplung des Makrolidfragments mit den Seitenketten. Diese Totalsynthese bestätigt die absolute Struktur und bietet einen Zugang zu analogen Verbindungen.
