As part of a program to develop low molecular weight thrombin inhibitors, the replacement of the dipeptide sequence (R)-Phe-Pro by mimetics is anticipated. Thus concise routes to (III), direct precursor of type (IV), and to the diastereomeric analogues (VI) and (VII) are developed.
