ChemInform Abstract

The solid phase synthesis of oligonucleotides containing 2′-deoxy-5-methylisocytidine and 2′-deoxyisoguanosine, which are of interest in DNA-based diagnostics, is presented. Various protecting groups for the phosphoramidates of these nucleotides in automated synthesis are evaluated. The N,N-diisobutylformamidine moiety is found to be a superior protecting group for 2′-deoxy-5-methylisocytidine.
