Abstract

Diethyl 2-amino-6-methylpyridine-3,4-dicarboxylate (I) is cyclized with the isothiocyanates (II) to yield the thioxopyrido(2,3-d)pyrimidinecarboxylates (III).

Diethyl 2-amino-6-methylpyridine-3,4-dicarboxylate (I) is cyclized with the isothiocyanates (II) to yield the thioxopyrido(2,3-d)pyrimidinecarboxylates (III). These are S-alkylated to give the alkylthiopyrido(2,3-d)pyrimidine-5-carboxylates (V). The analogous pyrido(2,3-d)pyrimidine-6-carboxylates (VII) are obtained by S-alkylation of (VI). (Vb), (Vc), and (VIIb) inhibit the aggressiveness of isolated animals and show analgesic activity.
